Every run in Tower Defense Simulator starts with the same question. You have a loadout of five towers, and there are 83 to choose from.
Some cost coins, some cost gems, some want Robux, and a surprising number are not for sale at all. You unlock those by beating a particular map.

There are 83 towers across 8 tiers. Only 2 are free, and 11 can no longer be unlocked at all.
Buying every tower that has a price comes to 169,600 coins and 32,000 gems.
The one that saves you Robux. Five towers let you either grind coins or pay Robux, and the deal is not the same. Gatling Gun gives you 18.0 coins of value per Robux, while Warden gives only 8.9. That is a 2.02 times difference for spending the same money.
How towers are grouped
The game sorts towers into tiers. Early tiers are cheap and simple, later ones cost more and do more specific jobs.

Plan this one early. Levelling a tower to 20 takes a long time. If you want Juggernaut, put Minigunner in your loadout now rather than later.
Towers you cannot buy (22)
These have no price at all. You get them by beating a specific map or finishing an event, so no amount of coins or gems will unlock them.
Event ones come back when the event does, so a missed one is not always gone for good.

The Robux price is not a fixed rate
Five towers can be bought with coins or with Robux. If the game used one exchange rate, every row below would show the same number. It does not.

Gatling Gun is the best deal here and Warden the worst, a gap of 2.02 times.
So if you do spend Robux, spend it on the expensive towers and grind the cheap ones.
Most towers are not bought at all
Only 31 towers have a coin price and 8 have a gem price. That leaves a large group you unlock by playing specific content instead.
So a big coin pile does not finish your collection. Beating maps does.
Free means two towers
You start with Scout and Sniper and nothing else. Every other tower in the game asks for something.
One tower in eight is gone
11 of 83 sit in the unavailable list, which is about 13% of everything in the game.
That is worth knowing before you chase one you saw in a video.

Is it worth buying towers with Robux?
It depends which one. Gatling Gun works out at 18.0 coins of value per Robux and Warden at only 8.9. The expensive towers are the better use of Robux.
How do I unlock Juggernaut, Kingpin or Operator?
Each needs a different tower at level 20 first, then gems and coins. Juggernaut needs Minigunner, Kingpin needs Crook Boss and Operator needs Scout.
Why can I not buy some towers?
22 towers unlock by beating a map or an event rather than by paying. Another 11 are no longer available at all.
What are golden towers?
Stronger golden versions of towers you already have. There are 8 of them and they come from golden crates.
